Visiting his animal friends at the Rio de Janeiro zoo, Tarzan learns that a tribal chieftain known as Barcuna has revived the ancient cult of leopard men and has been raiding villages along the Amazon and forcing the natives into slave labor. When his professor friend at the zoo is murdered by the leopard men, Tarzan sets out with Cheetah, his pet chimp, and Baron, his pet lion, to find Barcuna. Upon reaching the river, he encounters Capt. Sam Bishop, the owner of a small cargo boat, and his young aide, Pepe, an orphan of the Marakeet tribe. Captain Sam agrees to take Tarzan upstream as far as Calebah; but before reaching their destination they find Dr. Ann Phillips wandering about in shock following a raid by the leopard men. After being revived, she explains that she must get medicine to the Marakeet country to check an epidemic. When they arrive, however, the chief, out of fear and superstition, refuses to allow Dr. Phillips to inoculate his people. Young Pepe volunteers for the treatment; and when the natives see that no harm has befallen him, they all agree to accept inoculation. With the epidemic under control, Tarzan makes his way to Barcuna's hideout, where the captured slaves are being forced to dig for diamonds. The two men engage in hand-to-hand combat until Tarzan emerges triumphant. With the death of its leader, the leopard cult collapses. Captain Sam, Pepe, and Ann return to civilization, but Tarzan chooses to remain in the jungle for a while longer.

In the second week of filming Dinky the chimp bit 'Henry, Mike' on the jaw. It took 20 stitches to repair the wound. Henry had monkey fever delirium for three days, and took three weeks to recuperate. The chimp was destroyed and replaced.
